What is a monokaryotic culture? Its a culture that is grown for a single spore that hasn't been introduced to and/or paired with another spore.
So what can you do with a monokaryon? Because it hasn't mated with another spore you have the opportunity to select another strain for it to pair with and create a new strain.
How do you do that? Take a transfer from your mono and a transfer from a plate of another strain you want to cross it with and put them both on 1 plate. Where the 2 cultures meet on the plate you want to look for a spot that looks different than the growth from either of your transfers. A new, 3rd growth that is different from the growth from either of your transfers. And that is where you'll want to take 1 last transfer (or you can take multiple) and put that on a new plate by itself.
